Methods for determination of 2,4-dinitrophenol metabolites, namely 2-amino-4-nitrophenol and 4-amino-2-nitrophenol, were developed using HPLC with amperometric and spectrophotometric detection. Optimum conditions for the separation and determination were found, involving the mobile phase composition and detection potential.
The applicability of these methods for the determination of the analytes in model samples of urine was tested; solid-phase extraction was used for the preliminary separation from the matrix. This procedure allowed the determination of the analytes down to micromolar concentrations.
Amperometric detection proved to be more sensitive and selective than the spectrophotometric one.
