We investigated effect of metoprolol on bone metabolism in orchidectomised rats. Sham-operated control group (SHAM) and control group after orchidectomy (KON) received standard laboratory diet (SLD).
Experimental group after orchidectomy (MET) received SLD enriched with metoprolol for 12 weeks. Bone markers in serum-PINP, OPG, IGF-I, and in bone homogenate-CTX-I, BALP, BMP-2 were assessed.
BMD and biomechanical testing were performed. Bone markers (CTX-I, BALP, BMP-2) in KON were higher vs.
SHAM. In MET there was a decrease in CTX-I, PINP, BMP-2, OPG vs.
KON. In KON there was a decrease IGF-1 vs.
SHAM. In MET it was increased vs.
KON. Orchidectomy caused a significant decrease in BMD of whole body, lumbar vertebrae and left (LF) and right (RF) femur.
Biomechanical testing of LF and RF diaphysis revealed a decrease in maximal load values in KON vs. SHAM.
Metoprolol has a positive effect on bone metabolism in male rats by decreasing bone turnover and by increasing synthesis of IGF-1.
