Single- and Double-Headed Chemical Probes for Detection of Active Cathepsin D in a Cancer Cell Proteome

Abstract

We present the design of activity-based probes for cathepsins D that are effective in a complex proteome. The probes consist of a binding module with a statin-containing peptidic core, a warhead module bearing a photoreactive group, and a reporter tag.

The fluorescent and biotin tags make the probes a powerful tool for a wide range of "in-gel" and "on-blot" proteomic applications.
