Study of the damage and reapir of hepatocytes induced by xenobiotics in vivo and in vitro conditions

Abstract

This work was focused on the establishment of model injury to hepatocytes by tert butylhydroperoxide(tBHP),D-galactosamine(GalN)and thioacetamide(TAA)in vitro and on the evaluation of possible hepatoprotective action of S-adenosylmethionine in vitro(using these models)and in vivo(using TAA).Considerable damage to hepatocytes in primary cultures was achieved by model hepatotoxins using these concentrations and incubation periods-tBHP 1.5 mM and 30 min,GalN-40 mM and 24 h and TAA - 70 mM and 24 h.SAMe exerts protective effect on toxicity induced by model hepatotoxins in vitro.In spite of protective action of SAMe on damage in primary cultures neither the protective effect against liver injury induced by TAA nor the subsequent regenerative response of the liver in vivo were confirmed.
