The effect of high density lipoprotein on atherogenesis and increase in its concentration induced by CETP inhibiton

Abstract

Increased HDL-cholesterol is associated with decreased cardiovascular risk, but therapeutic possibilities to increase HLD-cholesterol are rather limited. A novel approach in this respect is represented by inhibition of CETP protein, which increases HDL cholesterol by 50% or even more.

However, torcetrapib - the first CETP inhibitor to be evaluated - substantially increased cardiovascular risk and total mortality. Mechanisms beyond these harmful effects remain obscure, and the future perspective of CETP inhibitors remains therefore uncertain.
