Oxytocin analogs for visualization of oxyticin receptors

Abstract

Nonapetid oxytocin (OT) is a hormone with peripheral and central effects. OT operates through one type of oxytocin receptor (OTR) of a G-regulatory proteins.

Current research aims to uncover mechanisms of action of OT on the molecular and cellular level to the therapeutic use of this knowledge. The key issue is the knowledge of the location and density of OTR in individual organs and tissues.

OTR presence of a protein is determined by immunohistochemical methods using specific primary antibodies. This method is very demanding, not only technically, but also economically.

For this reason, we sought an alternative option to visualize the oxytocin receptor. The invention relates to newly developed synthesis of fluorescent derivatives of oxytocin, which are intended for visualization of oxytocin receptors in mammalian tissues with OT labeled with a fluorescent marker, without using the primary antibodies.

The invention is the preparation of chimeras, derivatives of OT conjugated green emitting fluorescent dye {2- [phenyl (2H-pyrrol-2-ylidene-κN) methyl] -1H-κN pyrrolato} bor (Bodipy) using 1,3-dipolar cycloaddition.
