Liquid chromatography-tandem mass spectrometry in chiral study of amlodipine biotransformation in rat hepatocytes

Abstract

Amlodipine is a chiral calcium channel blocker. Racemic mixture is used in clinical practice even though it is known that only S-amlodipine possess vasodilating properties. Chiral aspects of biotransformation were studied by incubation of amlodipine and its two individual enantiomers with rat hepatocytes.

Eight metabolites presented in the highest amount were identified and semiquantified.
